Effect of The Extract of Porcine Testis on The Function of Immune-Related Cells

Abstract
Water-soluble extract and glycosaminoglycans (GAGs) were isolated from the porcine testis (PT), and their immuno-modulating activity was investigated. From an anion exchange chromatography used for further isolation of porcine testis GAGs (PT-GAGs), two fractions (PT-GAG-1.5 and PT-GAG-16) eluted by different salt concentration were obtained. In immunomodulating assays, water-soluble extract of PT (PT-Sol) showed the activity to suppress both concanavalin A (Con A)- and anti-CD3-mediated Tcell activation. However, PT-GAGs had no effect on lymphocyte activation. In and analysis for macrophage activation, PT-GAG-1.5, but nelther PT-GAG-16 nor PT-Sol, significantly enhanced the growth of macrophages. In addition to macrophage activation, PT-GAG-1.5 showed the activity to produce various cytokines such as interleukin-1¥â (IL-1¥â), interferon-¥ã (IFN-¥ã) and tumor necrosis factor-¥á (TNF-¥á) from murine microphages. These results suggest that PT possesses biologically active compounds to modulate the fuction of immune-related cells.
